Overview
Study: Ashley et al., Clinical Cancer Research, 2023. Memorial Sloan Kettering Cancer Center.
Cohort: 44 patients with newly diagnosed endometrial cancer, enrolled prospectively. Blood was drawn at baseline, after surgery, and every six months over 24 months.
Platform: Primary tumors were sequenced with a 468-gene targeted panel. Cell-free DNA was sequenced with a high-depth, 129-gene assay that uses molecular barcoding.
Findings: cfDNA levels correlated with surgical stage and were higher in advanced-stage disease. Before surgery, tumor mutations were detected in cfDNA in 8 of 36 patients with sequencing data, all with advanced-stage disease, high tumor volume or an aggressive histologic type. Circulating tumor DNA at baseline or after surgery was associated with shorter progression-free survival. In the 6 patients whose cancer recurred, changes in circulating tumor DNA mirrored progression and response to therapy, and in two cases came before clinically detected recurrence.
Data: The cBioPortal release (study ucec_ccr_cfdna_msk_2022) holds the primary-tumor sequencing: somatic mutations from 44 tumors (mutations called in 42), with histologic subtype, stage, lymphovascular space invasion, tumor mutational burden and, for 25 tumors, grade. The serial cfDNA results are reported in the paper, not in the release. Explore the cohort in the interactive atlas.
